At the end of the course the student will know the fundamental concepts of general chemistry, which are needed in order to address different professional fields, the basic principles of organic chemistry, the chemistry of functional groups, as well as the main reaction mechanisms of organic compounds. The student will also be aware of the structure and properties of biomolecules involved in the storage, transmission and expression of genetic information and the molecular mechanisms that underlie these processes. The knowledge of the fundamentals of molecular biology will allow the student to understand the biotechnological applications of this discipline and the resulting impact on medicine and diagnostics.
Chemical structure of nucleotides. Chemical structure of DNA: base pairing, double helical structures (A,B,Z),
DNA denaturation.
Different types of RNA: mRNA, tRNA, rRNA, snRNA. Supercoiling of DNA and nucleoid organization.
DNA metabolism: DNA replication, DNA polymerases in prokaryotes and eukaryotes, DNA repair, DNA recombination.
RNA metabolism: transcription of prokaryotic genes, transcription of eukaryotic genes, RNA polymerases and promoters, transcription factors, RNA processing, the reverse transcriptase. Translation: the genetic code and the different steps of protein synthesis.
Recombinant DNA technology: introduction to molecular biology technologies, DNA cloning, restriction endonucleases, cloning vectors, isolation of specific DNA sequences, complementary DNA, genomic and cDNA libraries, polymerase chain reaction (PCR) and sequence-specific oligonucleotides probes. Recombinant DNA technology applications: expression of cloned genes, site-directed mutagenesis, fusion proteins, cloning strategies in plants. Recombinant proteins for therapy. Analysis of gene expression profiles.
